IntroductionByvirtueofitsemissionnature,fluorescenceisanextremelysensitivetechnique,ascompared,forexample,toinstrumentationinvolvingabsorption.
Youcanthinkofemissiontechniquesasanalogoustoweighingapennybyplacingitdirectlyonascale,whileabsorptiontechniquesarecomparabletoweighinganelephantwithandwithoutapennyonitsback,thensubtractingonevaluefromtheothertoobtaintheweightofthepenny.
Becauseoffluorescencesensitivity,instrumentstomeasuretheemittedlightfromtheUVthroughIRarewidelyemployedinareasofbiochemistryandtechnology,toprobemoleculardistancesaswellasthelocalenvironmentofstructuresandcells.

TheFluorolog-3(Figure2)isthemostadvancedinstrumentavailable.
Themostprominentfeatureismodularity,whichencouragesyoutodesignaninstrumentthatisidealforyouruniqueapplication.
Forexample,youcanadddoublemonochromatorstoachievehigherthroughputandbetterrejectionofbackgroundradiation,oryoucanaddaspectrographandCCDdetectortoobtaincompletespectrainamatterofmilliseconds(Figure3).
Youcanoperateintheultraviolet,visible,orinfraredbyselectingfromavarietyofgratingsanddetectors.
Youcanalsochoosetomountmultipledetectorsorsourcesthatcomplementyourapplication.
